β-淀粉樣蛋白(amyloid β-protein ，Aβ)由郵淀粉樣前體蛋白(β-amyloid precursor protein,APP)水解而來，由細(xì)胞分泌，在細(xì)胞基質(zhì)沉淀聚積后具有很強(qiáng)的神經(jīng)毒性作用。在病理?xiàng)l件下，AD患者腦內(nèi)β淀粉樣蛋白（Aβ）的產(chǎn)生是正常情況下的4～10倍，大量的Aβ聚積形成的神經(jīng)炎性斑（老年斑），老年斑的形成導(dǎo)致海馬內(nèi)大量細(xì)胞的死亡。
阿爾茨海默癥（Alzhemer's disease,AD）是一種以進(jìn)行性認(rèn)知功能減退抱愧記憶障礙和癡呆為特征的神經(jīng)變性疾病，主要的病理熱證包括神經(jīng)細(xì)胞外以β-淀粉樣蛋白（Aβ）沉積為核心形成的老年斑（SP），細(xì)胞內(nèi)以超磷酸化的Tau蛋白為核心形成的神經(jīng)纖維纏結(jié)（NFT）及神經(jīng)細(xì)胞突觸的丟失和腦wei縮。Aβ是一種生理性多肽物質(zhì)，在腦中不斷生成和講解、清楚。AD發(fā)病機(jī)制中最主要的理論是淀粉樣肽瀑布假說，即腦內(nèi)Aβ產(chǎn)生和消除的不平衡導(dǎo)致疾病發(fā)生，最終導(dǎo)致神經(jīng)元退行性變而引起癡呆。
